Lots of people heading east on Rt 90 from points west.
Be advised that you exit the NY State Thruway from the left lane at Albany. Have a map quest print out handy, it's easy to get on the wrong road.
I'll be heading east Thursday around noon. I get onto Rt 90 east of the Rochester exits.

Rt. 90 is full of tolls. Last year I took Rt90- I86- NY17- I81- I88 and i think the only toll I paid was in Albany. My $.02

Well, it's 280 miles from Buffalo to Albany. Should be the last four hours of your trip.

If you're in Buffalo at 9 am, that means you be passing Rochester around 10 am and Syracuse around 11 am. Hang out at Niagara Falls for an hour and you'll miss the Buffalo morning rush hour.
I usually leave when I can't fit anything else in the car, usually around 11 - 1. I'll PM you with my phone numbers.
