I have a 2000 GT with the v6. The air conditioner has to ports, one up front by the radiator, and one on the passenger side behind the engine. Which one do I connect the can of refrigerant to?

the lower port. It should have a blue cap.

First things first, is your system empty?
if empty or low it means you have a leak, you need to first find and fix
or have it fixed or your just wasting your $$ and the environment.

and the way to fid a leak is to fill it and find the leak so he still needs to know where to fill it. Or he could go somewhere and they charge more than a can of r-134a

Starting to get off topic so this is my last post till we hear from
the OP,
To rix's post,
not always effective by it's self....but when used in conjunction
with a electronic leak detector you have a great chance at finding
it..
the problem with dye's are(if you can really call it a problem) they could
take a long time of driving with the AC on to find the leak if it's a smaller leak,
by the time it gets through the system and into the leak,and if it's
in a real tight hid spot you may not see it..:)
i start with my sniffer first....

But few DIYer's have a sniffer, or access to one.
Since our original poster *seems* to be a newcomer and wanting to DIhimself, I thought I would give him a hint of how to find the leak.
Rather than have him just self-recharge the refrigerant then scratch his head wondering how to find the leak.
And if the leak is so minor that he can't find it this summer, then overall he's prolly best served to just add a can or two each season rather than pay $100+ for repairs/recharge.
Frankly, refrigerant problems are SUCH a PITA, and require special equipment, that I don't do them myself.
Mebbe change a compresor if I know that's the problem, then have a shop evacuate and fill the system.
For nearly 50 years, GM has had an outstanding worldwide reputation for quality refrigeration systems.
Other than vac leaks in the HVAC controls, I never had any refrigeration problems (here in AZ) with any of over 15 GM's among me and my family.

Thanks for all the info guys. It looks like the refrigerant is leaking out of the high pressure port because there is oil (not motor oil) on it. I will add a can of 134 and see how long it holds the charge.

before you add any go and get a new H/P port, it
should turn off there's wench flats to do so take it to your
AP store and get a new one,,easy fix..
